In order to get a job, an actor has to audition. Auditioning is a big part of our job so we should try to learn from these opportunities. I remember reading for one of the "thugs" in "Nurse Betty" and was very surprised when I found that they cast Morgan Freeman and Chris Rock.

The casting process is so subjective so all you can really do is to stay open to learning and not to berate yourself.

If you're right for the part and they want you then most likely they'll use you. I had an interesting experience where I was "up" for a film and a commercial at the same time, my agent and manager and the producers all worked together to get me in on both of them. At first neither wanted to budge but we found a way to make the schedule work.
